Anscombe was named on the Wales bench, but pulled out of the match late due to a rib injury.
The experienced 31-year-old flyhalf slotted the winning conversion as Wales claimed their first-ever win in South Africa, beating the Springboks in Bloemfontein in the second Test and levelling the series.
His place on the bench will be taken by Rhys Patchell, who will get his first minutes of the Incoming Tour if he comes on to the field.
